There would indeed be a difference between the pack construction methods unless the cells were identical in spec (resistance)
Assume the 2500mah cell has a resistance of 5 milliohms
Two of them in parallel would lower the resistance to 2.5 milliohms
Unless the single 5000mah cell had the same resistance they would offer a different discharge performance

Given the same construction, larger cells have lower resistance. Probably is some difference though, partially due to extra connections, aspect ratios of the cells, etc, but not 2:1.
Not sure, but I suspect the 1P might be
slightly better largely because of fewer connections.
