Yes, if it's too tight it will restrict the drivetrain, the amount of side-to-side play you need is really tiny though, just a tick.
When I set the pulleys I usually do it without a pinion on my motor so I can spin the transmission and feel for any binding or unusual noises as more often than not they are caused by the pulleys being too tight.
