It won't be possible to get a second coat of glue between the bead of the tire and the rim because the glue is going to bond really fast. The biggest thing I've found when gluing tires is to take your time. I glue a little part of the bead on one tire, then move to the next while the first is drying, then move to the third, etc until I have made it through all the tires that I am gluing and then come back to the first and do a little more and run back through the rest of them and repeat until all the tires are glued rather than gluing one tire completely then moving to the second tire and gluing it completely before moving to the third and so on. Its been working well. I also peel the tire back from the rim and then fit it back where it goes a few times before I start gluing so that I have a feel for how it is going to be when I do glue them.
