Spin the wheel / tire slowly. When it comes to a stop, the heaviest spot will naturally fall to the bottom. The weight goes directly opposite of that - at the top of the wheel.
When properly balanced, you can spin the wheel and it will stop at a different point each time (i.e. no "heavy" spot).
