If the bolt on one side will loosen it, then logic states that having it on the other side tightens it. It's not going to tighten or loosen depending on what side it's on. Just pick a side that's easiest for you to adjust it from.
You forgot about the spring tension preventing the screw from tightening when installed correctly. If it is installed incorrectly the spring tension and the rotation will cause the bolt to loosen up.
